Suspicious fire at Vallejo mayor's law office being investigated as arson

VALLEJO -- Vallejo police are treating a suspected arson fire at the law offices of Vallejo Mayor Osby Davis early Saturday morning as domestic terrorism and have asked for assistance from federal investigators, fire and police officials said Saturday.

A Vallejo police spokesman told ABC7 that they are asking for FBI and ATF assistance to investigate the fire that gutted Davis' front office on Tuolumne Street across from the Solano County Superior Court.

"We're investigating it as arson at this time," Vallejo police Lt. Sid De Jesus said. "We don't know for sure, but we suspect it's an arson case ... The cause and origin is yet to be determined, but because it's an office occupied by the mayor, we're looking at this from every investigative aspect that we can."

Reached by phone, Davis said that to describe himself as "upset is an understatement."

He said he was awakened at about 1:30 a.m. by a phone call from fire Chief Paige Meyer and knew it was going to be bad news.

"Any time the mayor gets a phone call in the wee hours of the morning, it's never good," Davis said. "I really don't know what to say right now."

Davis said he will carry on.

"This is a bump in the road, a temporary hurdle," he said. "And we'll get past it and continue to do what we need to do."

It's too early to name a potential motive for the suspected arson, De Jesus said.

"We're not jumping to conclusions," the lieutenant said. "Both

(Davis and Micheal Thompson) are high profile people. It could be a disgruntled client of either attorney that occupied that office." Thompson shares the office with Davis, and has been a judicial candidate.

Calling the blaze, which appears to have started in the front waiting room area, "very suspicious," Meyer said the single-story office building in the 400 block of Tuolumne Street, was about 50 percent involved when crews arrived shortly after 1:30 a.m. Saturday.

"We knocked it down within about 15 minutes, but the building looks like nearly a total loss," Meyer said. The contents, however, were mostly salvageable, he said.

This is the second incident in recent months in which the mayor has been the victim of a possible crime. On May 18, Davis' 11-year-old motorcycle was stolen from the City Hall parking lot.

De Jesus said police are asking anyone who may have been in the area at the time of the fire and think they may have seen something suspicious, to call the Vallejo police nonemergency number, 707-552-3285.

Gory past sounds death knell for building | Local | News | The ...

If heritage advocates don?t scream too loudly over the Queens Ave. building targeted by Shmuel Farhi?s wrecking ball, there?s likely a very good ? or, at least, very grisly ? reason.

City politicians will likely decide in November whether to let the real estate magnate raze the building at 199 Queens, near Richmond St., to build what he pledges will be a $50-million mixed-use tower.

Council?s heritage advisers have OK?d Farhi?s plan ? and one of London?s most passionate history buffs has no doubt there will be little fight to stop the demolition on heritage grounds.

Why?

Because a push to underscore the historic value would no doubt focus on its single most significant event: it was where officials in 1879 found the body of Kate Gardner, believed to have been the first victim of a budding serial killer some consider a Jack The Ripper suspect.

?It would be the height of poor taste? to promote that, said heritage activist Joe O?Neil. ?If . . . My Sister?s Place or something like that wanted to move in and name the house after Kate Gardner that would be different, but to just say this should be a heritage house because of a murder would be offensive.??

Gardner, a pregnant, 21-year-old Tecumseh House Hotel worker, was found dead behind the house at 199 Queens on May 3, 1879. Also backing onto that spot was the Dundas St. office of Dr. Thomas Neill Cream, an abortionist.

Though a London jury found Gardner died ?from the effects of chloroform,? they did not name a suspect. But many in London eyed Cream ? who skipped town for Chicago, where he committed at least one murder and was jailed in 1881.

The Ripper-Cream link (which O?Neil and many serious historians doubt) hinges on when the deadly doctor was released from jail. Official records say it was 1891, three years after the never-found Ripper stalked London, England?s Whitechapel area and killed five prostitutes.

Some wonder, though, if he got out earlier and was back in England in 1888 for the infamous killing spree many believe must have been committed by a doctor.

This much is certain: Cream was back in England in 1891 and 1892 and was convicted and hanged for killing four prostitutes. (He claimed ?I am Jack . . .? just as he was hanged.)

Angus McLaren, a University of Victoria professor who wrote about Cream?s England crimes, doubts he was the Ripper. But in a 2004 interview he told The Free Press: ?There was probably a copycat aspect to (Cream?s) killings in the sense that the notion of killing prostitutes must have been very much in the air? post-Ripper.

Back here in London, O?Neil says he could see no heritage advocate using the Gardner death to help preserve Farhi?s building at 199 Queens.

?I don?t know anyone in the past who has used an infamous murder to justify (saving) any sort of historic site.?

Farhi has pledged to build a $50-million structure that would include ground-floor commercial space, several stories of residential units and many levels of what he calls much-needed parking.

Songs in the key of sea

ScienceDaily (Sep. 28, 2012) ? Soft horns and a tinkling piano form the backbone of "Fifty Degrees North, Four Degrees West," a jazz number with two interesting twists: it has no composer and no actual musicians. Unless you count bacteria and other tiny microbes, that is.

The song is the brainchild of Peter Larsen, a biologist at the U.S. Department of Energy's Argonne National Laboratory. Larsen, it turns out, has no musical training at all; his interests run less towards the blues and more towards blue-green algae.

When faced with an avalanche of microbial data collected from samples taken from the western English Channel, Larsen recognized he needed a way to make sense of it all. "Thinking of interesting ways to highlight interactions within data is part of my daily job," he said. "I am always trying to find new ways to visualize those relationships in ways so that someone can make relevant biological conclusions."

In the case of the western English Channel data, however, Larsen decided that a visual representation of the data would not be as effective as one he could hear.

"There are certain parameters like sunlight, temperature or the concentration of phosphorus in the water that give a kind of structure to the data and determine the microbial populations," he said. "This structure provides us with an intuitive way to use music to describe a wide range of natural phenomena."

A colleague of Larsen's suggested that classical music could effectively represent the data, but Larsen wanted any patterns inherent in the information to emerge naturally and not to be imposed from without.

"For something as structured as classical music, there's an insufficient amount of structure that you can infer without having to tweak the result to fit what you perceive it should sound like," Larsen said. "We didn't want to do that."

While this is not the first attempt to "sonify" data, it is one of the more mellifluous examples of the genre. "We were astounded by just how musical it sounded," Larsen said. "A large majority of attempts to converting linear data into sound succeed, but they really don't obey the dictates of music -- meter, tempo, harmony. To see these things in natural phenomena and to describe them was a wonderful surprise."

According to Larsen, the musicality of the data is not limited to the organisms in the English Channel. In another set of analysis, he and his colleagues used a similar methodology to look at the relationship between a plant and a fungus.

"We expect to see the same intuitive patterns recurring in different environments," he said. "Sometimes, it can sound a little avant-garde, but it's not random because it reflects very real phenomena."

Risk markers for erectile dysfunction following radiation treatment in prostate cancer

ScienceDaily (Sep. 27, 2012) ? In the first study of its kind, a research team led by Mount Sinai School of Medicine and Albert Einstein College of Medicine of Yeshiva University discovered 12 genetic markers associated with the development of erectile dysfunction (ED) in prostate cancer patients who were treated with radiation.

The findings, to be published online Sept. 27, 2012, in advance of the October 1, 2012 print issue in the International Journal of Radiation Oncology? Biology? Physics, the official scientific journal of the American Society for Radiation Oncology, are an important step towards helping clinicians determine the best course of treatment for prostate cancer patients and may lead to the development of therapies that alleviate side effects.

The main treatments for prostate cancer -- surgery, brachytherapy (seed implants) and external beam radiation therapy -- are all very effective at curing prostate cancer. Unfortunately, each treatment places patients at risk for ED. According to the National Cancer Institute, the prevalence of erectile dysfunction following external beam radiation for prostate cancer ranges from 65 percent to 85 percent. The Prostate Cancer Foundation estimates prevalence of ED following seed therapy at 25 to 50 percent. Many men will be able to regain their potency with time and treatments, but doctors would like to identify which men may be more likely to develop this side effect.

In the first large scale Genome-Wide Association Study to identify single nucleotide polymorphisms (SNPs) associated with susceptibility for the development of erectile dysfunction following radiotherapy for prostate cancer, researchers conducted a two-part study, first, to discover the candidate genetic markers of side effect risk, and second, to confirm which of those markers are replicated in a second group of patients. In the first group of prostate cancer patients, which included 132 men who developed erectile dysfunction after radiotherapy and 103 men similarly treated who did not develop erectile dysfunction, they found a set of genetic markers associated with erectile dysfunction. In the second part of the study, which examined 128 patients who developed erectile dysfunction after radiotherapy and 102 who did not, researchers confirmed that 12 SNPs were associated with erectile dysfunction.

"Thankfully, current treatments for prostate cancer offer excellent rates of long-term survival, so patients and their physicians have a choice about which treatment path to take," said Barry Rosenstein, PhD, Professor of Radiation Oncology, Mount Sinai School of Medicine. "However, the risk of developing erectile dysfunction after radiation treatment is highly variable, suggesting there may be a genetic component to determining that risk. Our study confirms that specific markers make certain patients more susceptible to this side effect."

Patients in the study cohort were given one of three treatments: internal radiotherapy, known as brachytherapy; brachytherapy plus external beam radiation; or external beam radiation alone. They were followed for an average length of nearly four years to determine level of sexual function after treatment.

Interestingly, the 12 SNPs identified in this study were located near genes that seem to be related to erectile function rather than related to radiation response. The researchers conclude that these SNPs may affect genes that sensitize a patient to developing erectile dysfunction when exposed to radiation during therapy.

"Prostate cancer screening and treatment are undergoing major shifts," said Harry Ostrer, MD, Professor of Pathology and Genetics at Albert Einstein College of Medicine and Director of Genetic and Genomic Testing at Montefiore Medical Center and co-principal investigator of the study. "This is part of our ongoing effort to identify men at highest risk for disease, identify the aggressive tumors that would be responsive to therapy, and to improve quality of life for men with indolent prostate cancers who might benefit from active surveillance, rather than therapy."

The authors indicate that examination of a large, independent cohort of similarly treated patients will be necessary to definitively determine which SNPs to include as part of a clinically useful predictive test to identify which men are at greatest risk for developing erectile dysfunction following prostate cancer radiotherapy. The researchers are also evaluating the impact of radiation treatment on urinary complications and proctitis, the inflammation of the rectum.

This study was supported by the American Cancer Society, United States Department of Defense, and the National Institutes of Health.

RIM buys itself breathing room for BB10 launch

TORONTO (Reuters) - Research In Motion Ltd shares jumped on Friday after the embattled BlackBerry maker posted quarterly results that showed it was still able to pull off a surprise as it tackles the formidable task of getting consumers excited over its new smartphone line.

While RIM's performance gave Wall Street a modicum of optimism, analysts stressed RIM has to now prove that the BlackBerry 10 devices, due early next year, can halt its brand's downward spiral. That won't be easy, they said.

"I have to admit they did a very good job making the best out of a really tough situation. You've got to give management some credit for that, but they still need to nurse the company back to health," said Sterne Agee analyst Shaw Wu.

RIM stunned investors on Thursday with stronger-than-expected BlackBerry sales in the quarter ended September 1. It even managed to build up its cash reserves, giving it a fighting chance to market the new BlackBerry 10 line effectively, while ramping up production of the reengineered smartphones.

Investors responded to the rare glimmer of good news for the embattled company by pushing up RIM's downtrodden share price up by more than 8 percent in afternoon trading on Friday.

Until Thursday's results, RIM was being written off by analysts because of its failure to keep pace with innovations from rivals such as Apple Inc and Samsung Electronics Co.

Analysts said RIM needs to generate enough excitment about the new devices to be able to sell them at a healthy profit.

"The question is, even if they do sell BB10 units, what is the margin profile going to be," said Wu, who notes that RIM once realized gross margins in the mid-40 percent range. Margins are now below 30 percent.

"If the gross margin profile stays where it is, then it is going to be very tough to be profitable, even if they sell a lot of BlackBerry 10 phones," he said.

To be sure, RIM managed to boost average selling prices in its latest quarter compared with the previous three months. That was a positive signal in the lead-up to the BB10 launch in early 2013.

"The bad news is that these results have little or no bearing on what remains true, and that is, RIM still needs to execute on BB10 and do it in a way that draws clear and resonant distinctions between what it has to offer and what the rest of the market has to offer," said CCS Insight analyst John Jackson.

The company is counting on BB10, equipped with a revamped operating system, to arrest a precipitous decline in market share over the past year and longer.

But in the end, the success or failure of the BB10 will hinge by how warmly it's embraced by consumers, many of whom have already switched to high-end devices like Apple's iPhone and Samsung's Galaxy S III, especially in North America and Europe.

"In the big picture, nobody, not even RIM knows whether this is going to work for them," cautioned Charter Equity Research analyst Ed Snyder.

Test versions of the BB10 have won praise from carriers and developers, but the true test still hinges on RIM's ability to win over fans of touchscreen devices.

"I want to regain market share in the U.S. and I want to win back market share in the full touch segment - that's what the company is geared up to and that's what we are working toward," said RIM Chief Executive Thorsten Heins in an interview with CNBC on Friday.

RIM's second-quarter performance also reminded investors that the company still enjoys a strong franchise in emerging markets, where consumers have graviated to its reliable though outmoded legacy smartphones.

Even though RIM's aging line-up has lost ground in North America and Europe, the BlackBerry maker managed to top shipment and revenue expectations in its second quarter, thanks largely to strong sales of smartphones in countries like India and Indonesia.

"This performance is nothing short of shocking as RIM has found a formula to entice its global carrier customers to sell (its) product," said National Bank Financial's Kris Thompson.

On Friday National Bank Financial upgraded RIM's stock, while BMO Capital Markets and Barclays Capital were among brokerages that raised price targets on the company's shares.

Thompson, who is rated five stars by Thomson Reuters StarMine for the accuracy of his estimates on RIM's earnings, upgraded the stock to "outperform" and increased his price target on the stock to $12 from $8.

Shares of RIM, which rose 20 percent after the closing bell on Thursday, were up 8.5 percent at $7.75 in afternoon trading on the Nasdaq on Friday. Its Toronto-listed shares rose more than 9 percent to C$7.57.

Police investigate a shooting at Accent Signage Systems on the north side of Minneapolis Thursday, Sept. 27, 2012, that left at least two people dead and four others wounded.

Two people were killed and at least four others wounded Thursday in a workplace shooting at a sign company in Minneapolis, according to local news reports. The gunman, an employee who had been fired earlier in the day, was among the dead, Fox9 reported.

Police arrived at the Accent Signage Systems Inc. on Chestnut Ave. West in the Bry Mawr area at approximately 4:45 p.m. to find the door propped open and several shooting victims inside, the station reported.


Fox9 said two people were dead after the shooting, including the gunman, and three shooting victims were transported to Hennepin County Medical Center in critical condition.?

Minneapolis police spokesman Steve McCarty did not confirm the death toll, saying only that there were "a couple of fatalities" and?at least four people hospitalized,?according to the Associated Press.

The station quoted an unidentified high-ranking law enforcement source as saying that the gunman returned to the office after being terminated earlier in the day. The source also told Fox9 that the gunman, whose body was found in the basement, may have been targeting specific employees.

After the shooting, dozens of squad cars and police vehicles surrounded the business. Traffic was stopped on a nearby bridge along Penn Avenue, where law enforcement officers had rifles drawn and pointed at a park below. People from the neighborhood milled around but deputies kept them back.? ?

Marques Jones, 18 of Minneapolis, said he was outside a building down the street having his picture taken when he and his photographer heard gunfire that sounded close.?

"We heard about four to five gunshots," Jones said. "We were shocked at what happened and we just looked at each other. We all just took off running to our vehicles."

The Latest on "Honey Boo Boo" | Famecrawler

We'll be seeing more of Honey Boo Boo's family soon.

Here Comes Honey Boo Boo aired its season finale last night on TLC ? which should leave you feeling either disappointed or relieved, depending on which side of the Love It/Loathe It fence you?re on.

For the last few weeks, our national attention has been drawn to the antics of Alana ?Honey Boo Boo? Thompson and her unique family. Jimmy Kimmel even made a joke about the show in the first few minutes of the Emmys broadcast. Meanwhile, Honey Boo Boo and her mother ?Mama June? Shannon have become the hottest reality personalities in town.

So if you think we won?t be hearing about the family for a while now that the show is on hiatus, think again! Here?s what we?ve found out about Alana and her clan in just the last couple of days?

They?ll be home for the holidays. And on our home TVs, too. According to Us Weekly, TLC has ordered more episodes of the hit series, which will feature Honey Boo Boo, Mama June, Sugar Bear, Pumpkin, Chubbs and Chickadee celebrating Halloween, Thanksgiving and Christmas.

Sugar Bear is back home. After an ATV accident, Alana?s dad, Mike ?Sugar Bear? Thompson developed an infection in his leg that landed him in the hospital for nearly two weeks. It was thought that he might need surgery ? and possibly even require amputation. But the infection has cleared up with strong doses of antibiotics, and cameras caught him returning home yesterday.

Little Kaitlyn won?t be getting corrective surgery yet. On last night?s episode, June?s oldest daughter, Anna ?Chickadee? Shannon, gives birth to a baby girl. Viewers also learned that the baby has an unusual birth condition ? two thumbs on her right hand. But contrary to what you may think, the condition isn?t necessarily easy to correct. June recently explained that the extra appendage will be staying for a while. ?The doctor doesn?t want to do anything right now because they don?t know the full function of the other thumb or if they?re connected,? she says. Removing the thumb might cause Kaitlyn to lose function in the rest of her hand. ?They don?t want to make any rash decisions.?

The family is enjoying its fifteen minutes of fame. June bristles at the suggestion that she?s harming Alana and her sisters in any way by doing the show. ?I don?t exploit Alana,? she told RadarOnline. ?I would never exploit my kids. Critics are critics, and everyone has their opinions, but I am not doing anything wrong. It?s always been about fun, and it?s an experience in life they can look back at. We watch the show and wish they put in more stuff!?

They may be getting more money for the show. Holla?! According to an insider source, the family is negotiating a higher salary for a new season, and will probably end up receiving around $8,000 per episode. Still, that?s nowhere near the $30 million the Kardashian family reportedly is receiving for their next three seasons.

They?re not as unusual as you might think. Okay, not everyone tries owning a miniature pig or goes on the kiddie beauty-pageant circuit. But in one aspect, June and her daughters are very much like many of their fellow Georgia families: Nearly 40 percent of families in the state are at risk of health problems related to childhood obesity. A major Georgia pediatric hospital issued a statement: ?[B]ased on the children and families we see at Children?s Healthcare of Atlanta, the truth is that Honey Boo Boo and her family?s health habits may be more of the norm in Georgia than everyone would like to believe.?

Crisis Intervention Service
Background Information
In the ten Local Health Offices that compromise the former EHRA
(Eastern Health Board) Region the Crisis Intervention Service (CIS)
provides an out of hours emergency social work service to young people
aged under 18 years who are in crisis. The service was established in
1992 and originally responded to young people between the ages of 12
and 18 years. Since 2005 the CIS was expanded to include all children
up to the age of 18 years.

The service is compromised of:

- A day Social Work Team.

- An Emergency Social Work Service which is available from Monday to
Sunday between 6.00p.m. and 6.00a.m. and each Saturday/Sunday and
Public Holiday from 9.00a.m. to 5.00p.m. all year round.

- A Night Reception Centre which is provided in Lefroy House for young
people who regularly present to the Out of Hours service. The service
is available from 5.00p.m. to 11.00p.m. and provides one to one
support with meals and showers for young people. This service is also
available to young people who are not availing of the Out of Hours
service.

- Emergency Foster Care Families who are available to provide a place
of safety as required for three nights for children under the age of
12 years of age.

- Seven emergency residential beds available on a night by night basis
at Lefroy House, Eden Quay for young people aged between 12 to 17
years of age.

- One emergency residential bed available on a night by night basis at
Sherrard House Residential Unit, North Circular Road for young girls
aged between 12 to 17 years of age.

- Four emergency beds available for a period of two weeks with CIS
designated Foster Care/Supported Lodgings for young people aged
between 12 to 17 years of age.

- The Crisis Intervention Service Partnership (CISP) which is an
Outreach Service in partnership between the Health Service Executive
and Focus Ireland. It was developed to ensure that there is a
comprehensive range of services available to support young people out
of home or availing of emergency accommodation. The CISP team is
available 9.00am to 5.00pm Monday to Friday. The Partnership works
with the local area social work teams to monitor the risk and welfare
of the young person while out of home. The Partnership is the internal
co-ordinator within the CIS services to ensure that there is overall
capacity within the service so that it can respond to ongoing demands.
The service works up individual plans with each young person designed
to ensure their welfare is being monitored, and that they are linked
with the appropriate supports. The service works closely with Focus
Ireland Outreach to provide out of hours supports pending placement in
emergency accommodation.

Referrals are received to the Crisis Intervention Service by telephone
from service providers working at night, for example An Garda
Siochana, Hospitals, Ambulance Services. Referrals are accepted in
relation to:

-? Concerns regarding the immediate welfare of children.

-? Young people in crisis seeking emergency accommodation.

- Young people who are identified by the Garda National Immigration
Bureau as separated children seeking asylum.

The Out of Hours is an intervention service and provides an emergency
response at night. All details of contacts with children are passed to
the local social work team by the start of the working day. The local
social work team are the case managers and will follow up on any
further assessments or interventions necessary.

The remit of the Crisis Intervention Service is to respond to crisis
situations whereby a child or young person requires immediate
placement, either due to child protection concerns, where there is a
breakdown in family relationships, placement breakdowns or due to
accommodation issues. CIS aims to prevent children or young people
from having to access emergency care unnecessarily. Where it is
appropriate, CIS tries to place children or young people with
alternative family members or friends or mediate between
children/young people and parents where there is a breakdown in family
relations. The Crisis Intervention Service put a significant amount of
effort into ensuring that placing a child or young person within
emergency residential units or foster care is a last resort.

Summary of Admissions to Crisis Intervention Service 2011
During the year 2011, the total number of children who were
accommodated in CIS emergency placements was 288. This figure relates
to 109 children aged 12 years and under and 179 young people aged
12-17 years.

The breakdown for reasons for admission for both age categories is
outlined below.

Summary of Admissions to CIS from January- June 2012
During the 1st and 2nd quarter of 2012, a total of 141 children/young
people were referred to the CIS for emergency accommodation.

Between January-June 2012, 48 individual children aged 12 years and
younger were placed in CIS emergency foster placements. The reasons
for admissions/referrals are outlined below.

of age were placed in CIS emergency residential/foster care/supported
lodgings placements. The reasons for admissions/referrals are outlined

Conclusion:

-? As demonstrated in the above tables, the majority of referrals to
CIS emergency accommodation for young people aged 12-17 years of age
relate to family relationship breakdowns. This phrase relates to
situations whereby a parent/guardian is stating that they are refusing
to have their child return to the family home due to the young person
engaging in anti-social behaviour such as being under the influence of
substances unknown, being aggressive and involved in criminalised
behaviour.

- The young person is in need of immediate accommodation on the night
due to the parent/guardian?s decision not to let them return to their
home; so technically while are they are in need of emergency
accommodation, the young person and their parents are not presenting
as ?homeless? as their parents/guardians have a place of residency,
but are refusing access to their child. The CIS is clear that when
young people are presenting in such circumstances or where the primary
referral reason is that they are in need of accommodation due to other
factors that are not related to being ?homeless? they are classified
as either ?seeking accommodation? or ?family relationship breakdown?.

- The CIS is aware that the referencing of young people as ?seeking
accommodation? due to relationship/placement breakdowns is open to
differential interpretation.

- Where there is a young person whose primary reason for referral to
CIS is because they and their parents/guardians are homeless; they
have no accommodation or home address, then they are classified as
homeless in CIS statistics.

- No children aged 12 years of age and under have been referred to the
CIS during 2011 and so far during 2012 due to homelessness

Whether you bought a brand new iPhone 5 or you upgraded an existing device to iOS 6, one of the first concerns everyone has is battery life. Is it better? Is it worse? Is it the same? A new device or new version of iOS typically does more than the last one, and because it's new, we typically use it more, at least in the beginning. Problems with backups, with push, with location services, with apps that don't shut down properly all also seem more common as everyone, from Apple to developers, gets used to the new hardware and software.

When the "fire brigade" arrives too late Impaired protein degradation causes muscle diseases RUB researchers and international colleagues report in Brain

New insights into certain muscle diseases, the filaminopathies, are reported by an international research team led by Dr. Rudolf Andre Kley of the RUB's University Hospital Bergmannsheil in the journal Brain. The scientists from the Neuromuscular Centre Ruhrgebiet (headed by Prof. Matthias Vorgerd) at the Neurological University Clinic (Director: Prof. Martin Tegenthoff) cooperated with colleagues from eleven institutes in seven countries. Among other things they found that protection mechanisms to combat abnormal protein deposits do not work properly in filaminopathy patients. This opens up new starting points for therapies that the team aims to test on cell cultures.

How filaminopathies develop

Mutations in the filamin C gene (FLNC) cause filaminopathies, which are manifested through progressive muscle weakness to the point of loss of the ability to walk. Muscle fibres are composed of myofibrils, for the development and maintenance of which the protein filamin C is crucial. The mutations examined in the study bring about a so-called myofibrillar myopathy: the myofibrils disintegrate in certain places and mutant filamin C and other proteins aggregate massively in the muscle fibres.

Support of protein degradation does not start on time

The researchers showed that the diseased protein deposits interfere with the protein degradation usually occurring in cells. Normally, cells produce what are known as heat shock proteins, which promote the degradation of protein deposits and make sure that other proteins assume their correct three-dimensional structure. "However, these protection mechanisms only seem to be increasingly activated when the critical point is exceeded. It looks as if the 'fire brigade' was called too late", says Dr. Kley. "We hope to positively influence the course of the disease by means of early treatment with substances that stimulate the production of heat shock proteins or affect the protein degradation in other ways. To study this, we have developed a cell culture model that allows us to carry out the first therapy studies in the laboratory."

Clinical picture more precisely characterised

The study of filaminopathy patients also enables the researchers to describe the disease more accurately now. The heart is more affected by the disease than previously thought, which may cause sudden cardiac death. It was also confirmed that pathological remodelling processes in the leg muscles conform to a specific pattern, which is visible on magnetic resonance imaging pictures. "This enables us to distinguish filaminopathies from other muscle diseases within the group of myofibrillar myopathies", explains Dr. Kley.

When the "fire brigade" arrives too late Impaired protein degradation causes muscle diseases RUB researchers and international colleagues report in Brain

New insights into certain muscle diseases, the filaminopathies, are reported by an international research team led by Dr. Rudolf Andre Kley of the RUB's University Hospital Bergmannsheil in the journal Brain. The scientists from the Neuromuscular Centre Ruhrgebiet (headed by Prof. Matthias Vorgerd) at the Neurological University Clinic (Director: Prof. Martin Tegenthoff) cooperated with colleagues from eleven institutes in seven countries. Among other things they found that protection mechanisms to combat abnormal protein deposits do not work properly in filaminopathy patients. This opens up new starting points for therapies that the team aims to test on cell cultures.

How filaminopathies develop

Mutations in the filamin C gene (FLNC) cause filaminopathies, which are manifested through progressive muscle weakness to the point of loss of the ability to walk. Muscle fibres are composed of myofibrils, for the development and maintenance of which the protein filamin C is crucial. The mutations examined in the study bring about a so-called myofibrillar myopathy: the myofibrils disintegrate in certain places and mutant filamin C and other proteins aggregate massively in the muscle fibres.

Support of protein degradation does not start on time

The researchers showed that the diseased protein deposits interfere with the protein degradation usually occurring in cells. Normally, cells produce what are known as heat shock proteins, which promote the degradation of protein deposits and make sure that other proteins assume their correct three-dimensional structure. "However, these protection mechanisms only seem to be increasingly activated when the critical point is exceeded. It looks as if the 'fire brigade' was called too late", says Dr. Kley. "We hope to positively influence the course of the disease by means of early treatment with substances that stimulate the production of heat shock proteins or affect the protein degradation in other ways. To study this, we have developed a cell culture model that allows us to carry out the first therapy studies in the laboratory."

Clinical picture more precisely characterised

The study of filaminopathy patients also enables the researchers to describe the disease more accurately now. The heart is more affected by the disease than previously thought, which may cause sudden cardiac death. It was also confirmed that pathological remodelling processes in the leg muscles conform to a specific pattern, which is visible on magnetic resonance imaging pictures. "This enables us to distinguish filaminopathies from other muscle diseases within the group of myofibrillar myopathies", explains Dr. Kley.
